Antibody humanization service

This service is provided on the back of over 15 years of antibody engineering and humanization experience, including the humanization of almost 150 antibodies, some of which have progressed into the clinic. We have completed projects for antibodies from mouse, rat, hamster, rabbit, llama, chicken and non-human primate with a success rate of >98%. 


This wealth of in-depth experience has enabled us to establish a low cost and very rapid service for the in silico design of humanized sequences. From a starting non-human sequence we will provide a panel of humanized sequences within 48 hours for $2000, including an assessment of sequence liabilities.

Humanization design process

Our humanization process, like most, is based on the concept of grafting CDRs from the parental non-human antibody onto a human framework.  Specifically we cycle through the following steps:

  1. Selection of CDRs based on a bespoke CDR definition that is a mix of Kabat, Chothia and IMGT.
  2. Select one or more homologous human frameworks from a curated subset of human germlines.
  3. Sequences regions outside of the CDRs that are known to impact binding directly or indirectly (e.g. Vernier zone) are analysed and where required back mutations are incorporated to try and ensure full antigen binding affinity and specificity is retained.


Throughout this process sequences are analysed for liabilities, such as glycosylation motifs, deamidation sites etc. Depending on the location of these motifs they may be removed or at least highlighted to the client for potential further investigation.

Deliverables

Specific deliverables are dependent on the finer details of the project, which will be agreed in advance. In general though all humanization projects will deliver the following:

  • A minimum of four humanized VH and four humanized VL sequences, providing 16 potential humanized antibodies. Some projects may give as many as 25 humanized antibodies depending on the starting sequence.
  • Analysis, and potentially removal of, sequence liabilities.
  • Full report containing assignment of CDRs by mulitple methods, humanized sequences, assessment of humanness, sequence liability analysis.


A complete report will be provided within 48 hours of receipt of an order.
